    Dihydroethidium简称DHE,是最常用的检测细胞内超氧化物阴离子水平的荧光探针。分子式为
C21H21N3,分子量为315.41。
    Dihydroethidium是一种最常用的超氧化物阴离子荧光检测探针。可以直接用于活细胞的标记。
Dihydroethidium被活细胞摄入后,可以在细胞内的超氧化物阴离子作用下脱氢,产生ethidium。Ethidium (例如溴化乙锭)可以和RNA或DNA结合产生红色荧光。当细胞内的超氧化物阴离子水平较高时,产生的ethidium较多,红色荧光就较强,反之则较弱。这样就可以用dihydroethidium进行超氧化物阴离子水平的检测。
    Dihydroethidium本身为蓝色荧光,最大激发波长为370nm,最大发射波长为420nm,脱氢后和RNA或DNA
结合产生红色荧光,最大激发波长为300nm,最大发射波长为610nm,实际观察时也可以使用535nm作为激发波长。
    Dihydroethidium可以溶解于DMSO,配制成适当的母液。
    用于细胞内超氧化物阴离子检测时,Dihydroethidium的常用浓度为1-10μM。通常用含有0.5-5μM的
Dihydroethidium的适当溶液和细胞一起在37℃孵育30分钟左右进行荧光探针装载,随后可以适当洗涤,接着可以直接用流式细胞仪或其它适当荧光检测仪器进行检测。
